Retatrutide (LY3437943) is a triple hormone receptor agonist developed by Eli Lilly that simultaneously acts on GLP-1, GIP, and glucagon receptors. In the Phase 3 TRIUMPH-1 clinical trial (May 2026), Retatrutide delivered the strongest published weight-reduction data for any medication to date.

Mechanism of action

By simultaneously activating three metabolic pathways, Retatrutide exerts a synergistic effect on energy balance, insulin sensitivity, and fat breakdown. In TRIUMPH-1, participants on the 12 mg dose lost on average 28.3 % of their body weight after 80 weeks — in participants with a BMI ≥35 even up to 30.3 % after 104 weeks. Even lower doses showed strong effects: 9 mg → 25.9 %, 4 mg → 19.0 %.

Reconstitution: Example calculation

To achieve a working concentration of 10 mg/ml, the 40 mg vial is reconstituted with 4 ml of sterile bacteriostatic water (BAC water).
